秀丽线虫RNAi文库能够提供 11,000 个RNAi克隆,每个克隆靶向一个单独的基因。该文库以C. elegans ORFeome 文库为基础,将ORF克隆载体转化至高产出pL4440-Dest-RNAi喂养载体上,且最终以大肠杆菌HT115(D3)甘油菌形式储存。该克隆纯化的质粒可直接作为体外合成双链RNA的模板,并可通过浸泡或注射的方式对线虫进行RNAi。

该文库包含超过6000个基因的预测启动子,可以方便地构建GFP报告基因线虫株,从而鉴定基因表达的时间和位置。这种线虫“启动子组”的潜在应用包括构建启动子-GFP融合体,用于获得蛋白质表达图谱和亚细胞定位,以及芯片和酵母单杂交分析。通过启动子与线虫ORF文库结合,还可用于组织特异性RNAi、亚细胞蛋白定位和异位(ectopic)表达研究。
